RESUMEN: La diversidad genética poblacional sin lugar a dudas es una herramienta indispensable en el desarrollo de huella genética, sin embargo las metodologías empleadas en el desarrollo de ésta hacen complicado el análisis de los resultados por lo cual en este trabajo se propone desarrollar huella genética mediante el desarrollo de curvas de disociación de alta resolución (HRM) la cual es una metodología de punta y altamente sensible, capaz de detectar cambios en un solo nucleótido, además se reduce el riesgo de contaminación ya que el análisis se realiza en un tubo de PCR convencional cerrado. En el presenta trabajo se analizaron nueve muestras pertenecientes a nueve delfines Tursiops truncatus, el análisis se realizó con seis marcadores microsatélites: TexVet1, TexVet2, TexVet4, TexVet9, D22 Y Ev37Mn, donde los marcadores, TexVet1, TexVet2, D22 Y Ev37Mn resultaron ser polimórficos, el iniciador TexVet4 no resultó ser polimórfico por lo cual no se recomienda utilizar en trabajos posteriores con este fin, el marcador TexVet9 contrario a lo reportado fue capaz de diferenciar entre individuos aunque no en la secuencia consenso del microsatélite si no, río arriba de la secuencia consenso se detectó un cambio en un solo nucleótido, lo cual confirma la especificidad del empleo de curvas de disociación de alta resolución, además de la capacidad de este marcador para diferenciar entre individuos y así poder emplearlo en el desarrollo de huella genética. Una de las principales ventajas del empleo de curvas de disociación fue, que una vez estandarizado el método se realizó una matriz de TM de acuerdo al patrón de repeticiones, por lo cual ya no fue necesario secuenciar todas las muestras para confirmar el número de repeticiones.
ABSTARCT: Population genetic diversity is undoubtedly an essential tool in the development of genetic fingerprinting, but the methodologies used in developing this make it difficult to analyze the results in this paper which aims to develop genetic fingerprinting by developing dissociation curves of high-resolution (HRM) which is a cutting-edge methodology and highly sensitive, capable of detecting single nucleotide changes also reduce the risk of contamination because the analysis is performed in a conventional PCR tube closed . In this paper we analyzed nine samples from nine dolphin Tursiops truncatus, the analysis was performed with six microsatellite markers: TexVet1, TexVet2, TexVet4, TexVet9, D22 and Ev37Mn, where markers, TexVet1, TexVet2, D22 and found to be polymorphic Ev37Mn , the initiator was not to be polymorphic TexVet4 therefore not recommended for use in subsequent work to this end, the score TexVet9 contrary to what was reported was able to differentiate between individuals but not in the consensus sequence of the microsatellite or not, upstream of the consensus sequence was detected a change in one nucleotide, confirming the specificity of the dissociation curves using high resolution, plus the ability of this marker to differentiate between individuals and thus be able to use it in the development of genetic fingerprinting. One of the main advantages of using dissociation curves was once standardized the method performed an array of TM according to the pattern of repeats, so it was no longer necessary to sequence all samples to confirm the number of repetitions.
