Abstract:
RESUMEN: El virus dengue (DENV) es una de las arbovirosis más importantes a nivel mundial. Aedes aegypti es el principal vector y transmisor del dengue y otros arbovirus hacia humanos a través de la picadura del mosquito. No existe tratamiento contra el virus del dengue y el control de la enfermedad se basa principalmente en el saneamiento de lugares de cría de larvas y en el control químico del vector, sin embargo, debido al continuo uso y exceso de insecticidas los mosquitos han desarrollado resistencia. Por lo que es importante buscar nuevas alternativas para controlar la población del vector del dengue y otras arbovirosis. La técnica de ARN de interferencia (ARNi) es una aproximación molecular que tiene mucho potencial para control de plagas clave debido a que se pueden bloquear genes necesarios para el desarrollo o supervivencia de los organismos plaga, mediante la introducción de RNA de doble cadena (dsRNA) exógeno que provoca la degradación del mRNA. Existen actualmente estudios con RNAi para el control de Ae. aegypti, sin embargo, la producción de dsRNA ha sido mediante transcripción in vitro, un proceso costoso y con una producción limitada. Por ello el presente trabajo tuvo como objetivo la construcción de una plataforma para la producción de dsRNA que resulte menos costosa y que pueda ser llevado a una mayor escala.
Para lograr el objetivo se clonaron los fragmentos de los genes Cyp302a1, Ache y F2 en los sitios de restricción HindIII y XhoI del vector de expresión pL4440. Células E. coli HT115 (DE3) fueron transformadas con las construcciones y posteriormente se llevó a cabo la inducción mediante la adición de IPTG de L4440_Cyp302a1, L4440_Ache, L4440_F2 (realizadas en este trabajo) y de L4440_Cyp314a1, L4440_ChsA (realizadas en un trabajo previo). También se realizó una transcripción in vitro usando el estuche comercial AmpliScribe™ T7 High Yield. En todas las inducciones se obtuvo dsRNA excepto en L4440_ChsA. Los tamaños de los dsRNA obtenidos tanto in vitro como in vivo fueron 420, 445, 511 y 403 pb respectivamente. Se logró la construcción de los sistemas de producción in vivo así como la obtención de dsRNA mediante este sistema
con una producción en promedio de 1.8 μg de dsRNA por 3 mL de cultivo celular con O.D₆₀₀ de 1 y, de manera in vitro, con un promedio de 20 μg/μL a partir de 1 μg de producto de PCR. El costo beneficio de producción de dsRNA mediante sistemas bacterianos es una alternativa económica y eficiente por lo que se debe buscar e implementar una estrategia para mejorar la producción.
ABSTRACT: At worldwide, the dengue virus (DENV) is the most important arbovirosis. Aedes aegypti is the main vector and transmitter of the dengue and anothers arbovirus towards humans through the mosquito bite. There is not exist treatment against the dengue virus, and the disease control is mainly based in the sanitation of breeding sites of larvae and the chemical control of the vector, however, due to continuous and excessive use of insecticides the mosquitoes have developed resistance. So it is important to search new alternatives to control the population of the dengue vector and anothers arbovirus. The technique of interference RNA (RNAi) is a molecular approach that has much potential for control of key pests, because genes can be blocked genes required for the development or survival of the pests through exogenous double-stranded RNA (ds-RNA), which causes degradation of the mRNA. Currently exist studies whit RNAi for the control of Ae. Aegypti however the production of dsRNA has been by in vitro transcription, expensive process and with a limited production. For this reason, the present work was aimed at building a platform for the production of dsRNA that is less costly and that can be taken to a larger scale.
To achieve the goal, were cloned the fragments of the genes Cyp302a1, Ache and F2 in the restriction sites HindIII and XhoI from the expression vector pL4440. E. coli HT115 cells (DE3) were transformed with the constructs and subsequently the induction was carried out by the addition of IPTG from L4440_Cyp302a1, L4440_Ache, L4440_F2 (performed in this work) and L4440_Cyp314a1, L4440_ChsA (performed in a previous work). Also performed an in vitro transcription using the AmpliScribe™ T7 high yield transcription kit. In all inductions dsRNA was obtained except in L4440_ChsA.
The sizes of the dsRNAs obtained both in vitro and in vivo were 42, 420, 445, 511 and 403 pb respectively. The construction of the in vivo production systems was achieved as well as the obtaining of dsRNA by this system with an average production of 1.8 μg of dsRNA per 3 mL of cell culture with O.D.₆₀₀ 1 and in vitro, with an average of 20 μg/μL from 1 μg of PCR product. The cost benefit of producing dsRNA by bacterial systems is
an economical and efficient alternative and it is necessary to search and implement a strategy to improve production.
