Abstract:
RESUMEN: La contaminación de suelos con hidrocarburos derivados de petroleo es una de las problemáticas ambientales de mayor interés, ya que estos compuestos están compuestos de sustancias ampliamente reconocidas como peligrosas tanto para la salud humana como para el ambiente, y presentan altos niveles de solubilidad en agua y gran capacidad de adhesión a las partículas del suelo (Rinaldi & Da Silva, 2011). En este trabajo de investigación se evaluó el efecto del disolvente conocido como thinner sobre la lombriz de tierra roja californiana (Eisenia foetida) y se determinó cuál es su concentración letal media (CL50). Asi mismo, se determinó el efecto sobre las dinámicas de mineralización de C y N en suelo de bosque, suelo salino y
vermicomposta, analizando CO2, NO3 , NO2 y NH4 y además se evaluó el efecto
de la concentración de materia orgánica adicional. De igual manera, se evaluó el efecto del thinner y la adición de materia orgánica sobre la población responsable del proceso de nitrificación, y finalmente se hizo el aislamiento e identificación de microorganismos con potencial para la degradación de hidrocarburos y con actividad nitrificante.
El thinner se evaluó por cromatografía de gases y la composición fue tolueno (263 mg/ml), 2 – butoxietanol (48 mg/ml), metanol (305 mg/ml), benceno (1.40 mg/ml) y mezcla de hexanos (70 mg/ml) y además provocó adelgazamiento, decoloración y fraccionamiento en los tejidos de las lombrices de tierra (An & Lee, 2008). El valor de CL50 correspondió a 12.46 ml de thinner por kg de suelo (bs) (3273.44 mg tolueno/kg suelo (bs) y 17.19 mg benceno/kg suelo (bs)). Se observó mayor producción de CO2 en los tratamientos con adición de thinner y thinner más adición de materia orgánica en comparación con los tratamientos control. Se encontró que el contenido de materia orgánica y la adición de materia orgánica adicional ejercen un efecto de retención sobre los componentes del thinner, observándose mayores concentraciones retenidas en los tratamientos con suelo de bosque y vermicomposta, y en todos los tratamientos con adición de materia orgánica adicional (Prokop et al. 2016). Se encontró que la adición de thinner y residuo de zanahoria más thinner en suelos y vermicomposta provocó el aumento y la acumulación de las concentraciones de NH + y NO - y por el contrario provocó la disminución en la concentración de NO -. En la vermicomposta, se encontró que la adición de thinner y materia orgánica adicional provocó el aumento en la concentración de población amonificante, nitritante y nitratante y de mesófilos aerobios, sin embargo, ante la acumulación de NH4 y NO2 , y la disminución en la concentración de NO3 , se concluyo que se favorecen los procesos de desnitrificación (Scott et al. 2014). De las dinámicas realizadas, se aislaron 23 microorganismos, de los cuales se identificaron 11 usando la plataforma EzTaxon (Chun et al. 2007), en su mayoría resultaron gram (+). De los microorganismos identificados, Arthrobacter rhombi, Microbacterium maritypicum, Glutamicibacter soli, Brachybacterium paraconglomeratum y Microbacterium esteraromaticum pertenecen al filo actinobacteria. Por su parte Pseudochrobactrum saccharolyticum, Ochrobactrum pituitosum y Alcaligenes faecalis subsp. Phenolicus pertenecen al filo de las proteobacterias. Bacillus wiedmannii, Bacillus cereus y Bacillus thuringiensis pertenecen al filo de los firmicutes. Estos microorganismos poseen actividad previamente reportada sobre el tratamiento de contaminantes orgánicos e inorgánicos, y con participación en la mineralización de nitrógeno tanto en el suelo como en ambientes acuáticos, los cuales podrán ser utilizados en trabajos posteriores para bioremediación.
ABSTRACT: Soil contamination with oil derived hydrocarbons is one of the environmental problems of greatest interest, since these compounds are formulated with substances widely recognized as dangerous, both for human health and for the environment and have high levels of water solubility and high adhesion capacity to soil particles.
In this research, the effect of the solvent known as thinner was evaluated on the californian red earthworm (Eisenia foetida) and its median - lethal concentration (LC50) was determined. Likewise, the effect on the dynamics of mineralization of C and N in forest soil, saline - alkaline soil and vermicompost were determinated, analyzing CO2, NO3 , NO2 and NH4 and also the effect of the concentration of additional organic matter was evaluated. Similarly, the effect of the thinner and the addition of organic matter on the population responsible for the nitrification process was evaluated, and finally the isolation and identification of microorganisms with potential for the degradation of hydrocarbons and with nitrifying activity was made. The thinner was evaluated by gas chromatography and the composition was toluene (263 mg / ml), 2-butoxyethanol (48 mg/ml), methanol (305 mg/ml), benzene (1.40 mg/ml) and hexanes mixture (70 mg/ml) and also caused thinning, discoloration and fractionation in the tissues of earthworms. The LC50 value corresponded to 12.46 ml of thinner per kg of soil (bs) (3273.44 mg toluene/kg soil (bs) and 17.19 mg benzene/kg soil (bs)). Higher CO2 production was observed in treatments with addition of thinner and thinner plus addition of organic matter compared to control treatments. It was found that the content of organic matter and the addition of additional organic matter exert a retention effect on the components of the thinner, observing higher concentrations retained in treatments with forest soil and vermicompost, and in all treatments with additional of organic matter. It was found that the addition of thinner and additional organic matter plus thinner in soils and vermicompost caused the increase and accumulation of NH + and NO - concentrations, and conversely caused the decrease in the concentration of NO3. In the vermicompost, it was found that the addition of thinner and additional organic matter caused the increase in the concentration of ammonifying, nitriting and nitrating population and of aerobic mesophiles; however, the accumulation of NH + and NO2 , and the decrease in the concentration of NO3 , it was concluded that the denitrification processes are favored. Of the dynamics performed, 23 microorganisms were isolated, of which 11 were identified using the EzTaxon platform, mostly resulting in gram (+). From the microorganisms identified Arthrobacter rhombi, Microbacterium maritypicum, Glutamicibacter soli, Brachybacterium paraconglomeratum and Microbacterium esteraromaticum belong to the phylum actinobacteria. On the other hand, Pseudochrobactrum saccharolyticum, Ochrobactrum pituitosum and Alcaligenes faecalis subsp. Phenolicus belong to the phylum of proteobacteria. Bacillus wiedmannii, Bacillus cereus and Bacillus thuringiensis belong to the phylum firmicutes. These microorganisms have previously reported activity on the treatment of organic and inorganic contaminants, and with participation in nitrogen mineralization both in the soil and in aquatic environments, which may be used in subsequent bioremediation processes.
