Abstract:
RESUMEN: Los efectos del hipertiroidismo sobre la estructura y funcionamiento de las arterias
de conductancia no han sido dilucidados. La activación por las hormonas tiroidea
de las vías de señalización PI3K/Akt y MAPK/ERK - vinculadas con crecimiento,
diferenciación, migración y proliferación celulares - ha sido descrita en las células
endoteliales y musculares lisas de las aortas ratas, pero la relación entre la
activación de estas vías y la remodelación aórtica es desconocida. Nuestro objetivo
fue examinar en función del tiempo, las modificaciones en la expresión de
las proteínas Akt y ERK1/2 (y sus formas fosforiladas, p-), relacionándolas
con los cambios histomorfométricos y de la función motora, de las aortas torácicas
aisladas de ratas con hipertiroidismo agudo. Este fue inducido en las ratas
mediante la administración de T3 (500 μg/kg/día, s.c.) por 1(T31d), 3(T33d) y 10
días (T310d). Los controles correspondientes recibieron vehículo (solución salina
alcalina, s.c.) por 1(V1d), 3(V3d) y 10 días (V10d). El incremento de T3 sérica
permitió confirmar el estado hipertiroideo en las ratas. En las aortas de los grupos
definidos, se caracterizaron las variaciones histomorfométricas, las respuestas
contráctiles inducidas por la fenilefrina (agonista adrenérgico α 1), y las
expresiones de Akt, p- Akt, ERK1, p-ERK1, ERK2 y p-EK2. Al compararse con
los controles temporales respectivos: (i) las aortas torácicas de las ratas T31d,
T33d y T310d manifestaron reducción del número de núcleos de las células
musculares lisas, desorganización de estos núcleos en relación con las láminas
elásticas, y pérdida de compactación y discontinuidades de las láminas elásticas; (ii) las aortas de los grupos T33d y T310d - pero no T31d - mostraron depresión de la
contractilidad inducida por la fenilefrina, independientemente del endotelio
funcional; (iii) la expresión de Akt aumentó en el grupo T310d, sin modificarse en
los grupos T33d y T31d; en cambio, p-Akt y p-Akt/Akt disminuyeron transitoriamente
en las aortas de las ratas T31d (esto es, la disminución no se manifestó en los
grupos T33d y T310d); y (iv) ERK1/2 y sus formas activas no fueron modificadas
en las aortas de las ratas T31d, T33d y T310d. Así, durante el tratamiento con T3,
el curso temporal de los cambios histomorfométricos y la expresión disminuida de
p-Akt fueron coincidentes el primer día; sin embargo, la transitoriedad de la
disminución de p-Akt no permitió correlacionarla con los cambios
histomorfométricos sostenidos hasta el décimo día. El incremento de Akt total al
décimo día, excluye una relación causal con la remodelación aórtica y la
depresión de la actividad contráctil de aparición más temprana, en las aortas
de ratas con hipertiroidismo agudo. En conclusión, el tratamiento con T3 a corto
plazo ocasionó remodelación de la pared aórtica y depresión de la respuesta
contráctil inducida por la activación de receptores adrenérgicos α 1,
independientemente del endotelio, sin la participación de las vías de
señalización que comprenden a las proteínas Akt y ERK1/2. The effects of hyperthyroidism on the structure and function of conductance arteries
have not been elucidated. Activation by thyroid hormones of the PI3K / Akt and
MAPK / ERK signaling pathways - linked to cell growth, differentiation, migration and
proliferation - has been described in the smooth vascular muscle and endothelial
cells of the aortas, its relationship with aortic remodeling is unknown. Our objective
was to examine, as a function of time, the modifications in the expression of Akt and
ERK1/2 proteins (and their phosphorylated forms, p-), relating them to the
histomorphometric and motor function changes of Isolated thoracic aortas from rats
with acute hyperthyroidism. Which was induced in the rats by daily subcutaneous
injection of T3 (500 μg/kg/día, s.c.) by 1(T31d), 3(T33d) and 10 days(T310d). The
corresponding controls received vehicle (alkaline saline solution, s.c.) for 1(V1d),
3(V3d) and 10 days(V10d). The increase of T3 in serum allowed to confirm
the hyperthyroid state in rats T31d, T33d and T310d. In the aortas of rats of the
defined groups, histomorphometric variations, contractile responses induced by
phenylephrine (α1-adrenergic agonist), and the expressions of Akt, p-Akt, ERK1,
p- ERK1, ERK2 and p- EK2 was characterized. Compared with the respective
temporal controls: (i) the thoracic aortas of the T31d, T33d and T310d rats showed
a reduction in the number of smooth muscle cell nucleus, disorganization of
these nucleus in relation to the elastic lamellas, and loss of compaction and
discontinuations of the elastic lamellas (ii). Aortas of groups T33d and T310d - but not T31d - showed contractile depression induced by phenylephrine,
regardless of functional endothelium (iii). Akt expression increased in the T310d
group, unmodified in the T33d and T31d groups; In contrast, p-Akt and p-Akt / Akt
decreased temporarily in the aortas of the rats of T31d (that is, the decrease did
not occur in the T33d and T310d groups). In conclusion, during the treatment with
T3, the temporal course of the histomorphometric changes and the decreased
expression of p-Akt were coincident on the first day; however, the transience
nature of the p-Akt decrease did not allow it to correlate with the histomorphometric
changes sustained until the tenth day. The increase of total Akt to the tenth day,
excludes a causal relation with the aortic remodeling and the depression of the
contracting activity of earlier occurrence, in the aortas of rats with acute
hyperthyroidism. ERK1 / 2 and its active forms were not modified precociously.
T3 caused remodeling of the aortic wall and depression of the contractile
response induced by the activation of α1 adrenergic receptors, independently
of the endothelium, without the involvement of signaling pathways comprising
the Akt and ERK1 / 2 proteins.
