Abstract:
RESUMEN: Objetivo: Evaluar las modificaciones en la expresión de los factores angiogénicos
(factor de crecimiento endotelial vascular, VEGF; factor de crecimiento de fibroblastos, FGF; factor inducible por hipoxia 1α, HIF1α) y la densidad capilar coronaria durante el desarrollo de hipertrofia miocárdica en los corazones de ratas Wistar con hipertiroidismo a corto plazo.
Materiales y Métodos: El hipertiroidismo se indujo mediante la administración subcutánea diaria de 3, 5, 3' triyodo-L-tironina (T3; 500 g/kg) por 1 (T31d), 3 (T33d)
y 10 (T310d) días. Los animales control fueron inyectados con el vehículo (1ml/kg) durante 1 (V1d), 3 (V3d) y 10 (V10d) días. Se evaluaron: (i) los valores séricos de las
hormonas tiroideas y la masa ventricular izquierda cardiaca, (ii) la densidad capilar y el área de sección transversal de los cardiomiocitos en los ventrículos izquierdos (VIs) mediante análisis histológico morfométrico, (iii) la función cardiaca con la preparación de Langendorff con flujo constante, (iv) los cambios en los niveles de
las proteínas HIF1α, VEGF y FGF en los VIs, mediante análisis de transferencia Western. Resultados: En comparación con sus respectivos controles temporales, los corazones aislados de las ratas T33d y T310d, pero no así de las T31d, desarrollaron incremento de la masa ventricular izquierda, acrecentamiento del área de sección transversal de los cardiomiocitos, y valores absolutos aumentados de +dP/dtmax y -
dP/dtmin (esto es, inotropismo y lusitropismo positivos). La presión desarrollada por el ventrículo izquierdo, la presión de perfusión coronaria, y la frecuencia cardiaca espontánea de los corazones aislados de las ratas T31d, T33d y T310d no fueron modificadas. Interesantemente, los estudios histológicos mostraron una tendencia (no significativa) de la densidad capilar a aumentar en los VIs de las ratas T31d,
T33d y T310d, en comparación correspondiente con los controles temporales; así, el
crecimiento capilar fue paralelo al crecimiento de los cardiomiocitos (i.e., no se observó rarefacción capilar). En los VIs de las ratas T31d, T33d y T310d, el HIF-1
permaneció inalterado al compararse con los correspondientes valores de los
controles. En los VIs de las ratas T33d, la expresión del FGF fue mayor que la del
control V3d, pero sus niveles fueron similares en los grupos T31d y T310d
comparados con los valores obtenidos en los corazones de los grupos V1d y V10d,
respectivamente. Notablemente, los niveles de expresión de VEGF aumentaron en los VIs de los grupos T31d y T33d, pero no en los del grupo T310d, en relación con
los controles temporales.
Conclusión: Durante la fase temprana de desarrollo del hipertiroidismo
experimental en la rata, de forma aproximadamente coincidente en el tiempo, se manifiestan: hipertrofia ventricular izquierda, inotropismo y lusitropismo positivos, y aumento de la densidad capilar. Este último, hipotéticamente, permite mantener la perfusión miocárdica fisiológica. Consideramos, asimismo, que el mecanismo para la angiogénesis comprende el aumento transitorio de la expresión de los factores VEGF y FGF, que acompaña parcialmente la manifestación del incremento de la densidad capilar en los VIs de las ratas tratadas con T3. La ausencia de cambios significativos en la expresión del HIF1, excluye a este factor como participante
causal de la angiogénesis de temprano desarrollo en los VIs de las ratas tratadas con T3 a corto plazo.
ABSTRACT: Objective: To assess the modifications of angiogenic factors expression (vascular endothelial growth factor, VEGF; fibroblast growth factor, FGF; hypoxia-inducible factor 1α, HIF1α) and coronary capillary density during the development of myocardial hypertrophy in hearts of Wistar rat with short-term hyperthyroidism. Materials and Methods: Hyperthyroidism was induced by daily subcutaneous administration of 3,5,3' triiodo-L-thyronine (T3; 500 g/kg) for 1 (T31d), 3 (T33d), and
10 (T310d) days. Control animals were injected with the vehicle (1ml/kg) for 1 (V1d),
3 (V3d), and 10 (V10d) days. They were evaluated: (i) serum values of thyroid hormones and cardiac left ventricular mass, (ii) capillary density and crosssectional area of cardiomyocytes in the left ventricles (LVs) through morphometric histological analysis, (iii) cardiac function with constant flow Langendorff preparation, (iv) changes in the levels of HIF1α, VEGF, and FGF proteins in LVs by
Western blot analysis.
Results: Compared to their respective time controls, hearts isolated from T33d and
T310d rats, but not from T31d, developed enlarged left ventricular mass, augmented
cross-sectional area of cardiomyocytes, and increased absolute values of +dP/dtmax and -dP/dtmin (that is, positive inotropism and lusitropism). Left ventricular pressure development, coronary perfusion pressure, and spontaneous heart rate of
the isolated hearts of T31d, T33d, and T310d, rats were not altered. Interestingly, histological studies showed a tendency (non-significant) for capillary density to increase in the left ventricles of T31d, T33d, and T310d rats, in corresponding comparison with temporal controls; thus, capillary growth was parallel to cardiomyocytes growth (i.e., capillary rarefaction was not observed). In the LVs of
T31d, T33d, and T310d rats, HIF-1remained unchanged when compared with the
corresponding control values. In the LVs of T33d rats, FGF expression was higher than that of V3d control, but their levels were similar in the T31d and T310d groups compared to the values obtained in hearts of V1d and V10d groups, respectively.
Notably, the levels of VEGF expression increased in the LVs of T31d and T33d
groups, but not in those of the T310d group, when compared with the temporal controls. Conclusion: During the early phase of development of experimental hyperthyroidism in the rat, roughly coinciding in time, were manifested: left
ventricular hypertrophy, positive inotropism and lusitropism, and increased capillary density. The latter, hypothetically, allows the physiological myocardial perfusion to be maintained. We consider, also, that the mechanism for angiogenesis includes the transient rise in the expression of VEGF and FGF, which partially accompanies the manifestation of the increase in capillary density in the LVs of rats treated with T3. The absence of significant changes in HIF1expression excludes this factor as a causal participant of early development angiogenesis in the LVs of short-term T3 treated rats.
