ABSTRACT: Currently, is a major interest for specific fragments of dietary proteins, called bioactive peptides, which possess biological activity improving human health (antihypertensive, antioxidant, anticancer, anticholesterolemic, antithrombotic, etc), additionally these
peptides regulate physiological processes besides their nutritional value. Since middle 80’s there’s been a massive demand for the elaboration of food formulas from vegetal protein sources. In the present investigation, hydrolysates were prepared from protein concentrates of Lima bean (Phaseolus lunatus) using two enzymatic systems, Alcalase 2.4L and Pepsin-Pancreatin respectively, through a experimental design 23, where the effect of three factors was evaluated (protein concentrate source: of ungerminated grain CP1 and germinated grain CP2, enzyme/substrate relation E/S: 1/50 and 1/10, hydrolysis time: 0.5 and 2 h for Alcalase; 1 and 3 h for Pepsin-Pancreatin) in respect to hydrolysis grade and inhibitory activity of the ACE-I. The hydrolysates prepared with Alcalase showed GH between 24.12% and 58.94% and an inhibitory activity (IC50 amount of protein required to inhibit the 50% of the ACE-I activity) amid 0.56-2.4 mg/mL. On the other hand, in the hydrolysates obtained with Pepsin-Pancreatin, the GH’s were minor (15.35-37.07%), but better results in regard to inhibitory activity (IC50: 0.25-0.69 mg/mL). In the Size Exclusion Chromatography of the hydrolysates H4, H7,
H12 and H15 with higher antihypertensive activity were collected fourteen peptidic fractions with inhibitory values of the ACE-I ranging from 8.65-45.9 % and molecular weights between 1-0.75 kDa. In the purification step by HPLC of the two most promising fractions F7-4 and F12-4 both from the CP1, peptidic fractions were collected with superior inhibition activity of the ACE-I. In F7-4 three notable fractions were collected (F7B-6, F7B-7 and F7B-8) with IC50 values varying between 1.7-3.8 ?g/mL; from F12-4, the superb fractions were F12C-3, F12C-4 and F12C-5 with IC50 in the range of 0.9-3.4 ?g/mL. In the Aminoacid Profile of these six new fractions the increase
of Pro, Trp, Met and Phe was evident, as well in the content of hydrophobic aminoacids.
RESUMEN: En la actualidad, existe un creciente interés por determinados fragmentos específicos de las proteínas de la dieta, denominados péptidos bioactivos, que poseen actividad biológica benéfica al organismo (antihipertensivos, antioxidantes, anticancerígenos, anticolesterolémicos, antitrombóticos, etc), además de regular procesos fisiológicos y de su valor nutrimental. Desde mediados de los años 80’s ha existido una demanda de fuentes proteínicas vegetales para la elaboración de formulaciones alimentarias,
sustituyendo a los hidrolizados de origen animal. En el presente trabajo de investigación se efectuaron hidrolizados a partir de concentrados proteínicos de frijol lima (Phaseolus lunatus) empleando dos sistemas enzimáticos, el primero con alcalasa 2.4L y el segundo con pepsina-pancreatina a través de un diseño experimental 23 donde se evaluó el efecto de tres factores (fuente de concentrado proteínico CP: de semilla sin germinar CP1 y de semilla germinada CP2, relación enzima/sustrato E/S: 1/50 y 1/10 y tiempo de hidrólisis: 0.5 y 2 h para alcalasa;1 y 3 h para pepsina-pancreatina) sobre el grado de hidrólisis GH y sobre la actividad inhibitoria de ECA-I. Los hidrolizados preparados con alcalasa tuvieron GH entre 24.12 y 58.94% y una actividad de inhibición(IC50: cantidad de proteína requerida para inhibir el 50% de la actividad de ECA-I) entre 0.56 y 2.4 mg/ml. En los hidrolizados obtenidos con pepsina-pancreatina, los GH fueron menores (15.35-37.07%), pero se obtuvieron mejores resultados de inhibición (IC50: 0.25-0.69 mg/ml). En la cromatografía por filtración en gel de los hidrolizados H4, H7, H12 y H15 con mayor actividad antihipertensiva se recolectaron catorce fracciones peptídicas con valores de inhibición de ECA-I entre 8.65-45.9% y con pesos moleculares entre 1 y 0.75kDa. En la purificación por HPLC de las dos fracciones más promisorias F7-4 y F12-4 ambos a partir de CP1, se recolectaron fracciones peptídicas con mayor poder de inhibitorio de ECA-I. En F7-4 se recolectaron tres fracciones promisorias (F7B-6, F7B-7 y F7B-8) con IC50 entre 1.7 y 3.8 g/ml; a partir de F12-4, las fracciones promisoras fueron F12C-3, F12C-4 y F12C-5 con IC50 entre 0.9 y 3.4 g/ml. En el perfil de aminoácidos de estas seis fracciones obtenidas fue evidente el incremento de prolina, triptófano, metionina y fenilalanina, así como también en el contenido de aminoácidos hidrofóbicos.
