RESUMEN: El presente trabajo muestra la importancia de los probióticos específicamente de Lactobacillus acidophilus en la salud del huésped, haciendo énfasis en preservar la viabilidad en un lugar específico (colon) con el uso de una matriz polimérica, prebióticos y un proceso de secado por aspersión como un método de protección ante efectos deletéreos. La matriz polimérica es una mezcla de carboximetilcelulosa (CM), pectina (P) y proteína aislada de suero lácteo (WPI), a la cual se le realizo una caracterización de mezclas binaras y una terciaria con y sin proceso de desnaturalización (D y SD), en base a curvas de secado determinando su energía de activación (Ea) así como microestructura de las películas generadas. Por otra parte este mismo análisis se realizó con la presencia de prebióticos en base a un diseño Simplex centroide. Una vez conocida las características de la película se procedió a realizar la activación y cinética de la bacteria probiótica (Lactobacillus acidophilus) para conocer el tiempo en el cual se encontraba en la fase estacionaria, el monitoreo de cinética se realizó por medio de pH, Densidad óptica, Numero más probable (NMP), y método de impedancia, dando como resultado que la fase de interés se encuentra a las 18 h, coincidiendo el resultado en los 4 métodos de medición anteriormente descritos. Una vez obtenidos los microorganismos en fase estacionaria se realizó la microencapsulación por secado por aspersión, continuando con pruebas de viabilidad después del secado, resistencia a fluidos gástricos e intestinales simulados, así como su microestructura de los polvos obtenidos. Los resultados de las Ea de las mezclas binarias y ternaria reflejan que las mezclas D exhiben mayor energía de activación (40.14 y 37.19 KJ/mol) comparada con las SD, la presencia de prebióticos proporciona una Ea de 38.7 KJ/mol reflejando para P0.00I3.00A0.00 y P2.25I1.50A0.00 reflejando que esta disminución se debe a las propiedades de los materiales y formación estructural de la microcapsula. En el efecto del secado, la sobrevivencia se monitoreo por NMP e impedancia reflejando que las muestras P0.76I2.00A2.38, P0.76I0.51A9.30 y el control (P0.76I0.51A9.30) tienen mayor capacidad de protección reportando 6.5x10^6 NMP de microorganismos, y una resistencia alta confirmando que a niveles altos de microorganismo este parámetro es disminuido. Por otra parte en la estabilidad a condiciones simuladas de fluido gástrico e intestinal, nos indica que las capsulas que proporcionaron mayor protección fueron: P2.25I0.00A7.00, P3.00I0.51A2.38 (2.5x107, 1.5x10^7 respectivamente) y P0.00I1.50A7.00(3.3x10^8 ), esto datos son sobresalientes ya que se encuentran entre 3 y 4 ciclos logarítmicos más con respecto a las otras fórmulas desarrolladas y dos ciclos mayores respecto al control (1.3x10^5 ). Con respecto a la microestructura encontramos que la mezcla con mayor viabilidad tiene una estructura en forma de hojuelas con bordes definidos y ligeramente suaves, su apariencia es ligeramente compacta favoreciendo la protección y liberación de bacterias probióticas. Con estos datos podemos concluir que el uso de prebióticos aunados a una matriz polimérica protegen la viabilidad de Lactobacillus acidophilus de las condiciones de estrés por el proceso de microencapsulación y tránsito a través del tracto gastrointestinal, asegurando que las bacterias sean liberadas en cantidad suficiente en un lugar específico (colon) proporcionando beneficios al huésped.
ABSTRACT: In this work it is shown the importance of the Lactobacillus acidophilus probiotic in the health of a host. It was emphasized the preserving, and its viability in a specific site (the colon) by means of a polymeric matrix, probiotics, and an aspersion drying process, considered as a protection technique against deleterious effects. The polymeric matrix is a mixture of carboxymetilcellulose (CM), pectin (P), and milky whey isolated protein (WPI). Binary and thirdnary mixtures were prepared with and without a desnaturalization process, (D and SD). According to the drying behavior, the activation energy was estimated (Ea). Furthermore, the microstructure of the films obtained was also studied. In addition, this same analysis was performed with the presence of probiotics considering a centroid simplex design. Once the characteristics of the films were known, the activation of the probiotic bacterium (Lactobacillus acidophilus) was realized, and its kinetics was studied. From the measurements it was possible to estimate the time in which the bacterium is found in a stationary phase. The kinetics of the bacterium was observed by measuring the pH, the optical density, the most probable number, and the electric impedance. The results indicate that the phase of interest is found at about 18 hours, as indicated by the four methods above mentioned. Once the micro-organisms were got in the stationary phase, the microencapsulation was performed by means of an aspersion drying. Going on with the viability proofs, after the drying, simulated intestine and gastric fluid resistance, as well as the microstructure of the powders got, was studied. The results of the activation energy of the binary and thirdnary mixtures show that the D mixtures have a higher activation energy (40.14 y 37.19 KJ/mol), when compared with the SD mixtures. The presence of probiotics led to an Ea of 38.7 KJ/mol for P0.00I3.00A0.00 and P2.25I1.50A0.00. The drop in the Ea is due to the properties of the materials as well as to the structural formation of the microcapsule. Concerning the drying effect, the survival was studied by NMP and impedance spectroscopy, showing that samples P0.76I2.00A2.38, P0.76I0.51A9.30 and P0.76I0.51A9.30 (the control) have the better protection capacity, reaching as much as 6.5x10^6 NMP micro-organisms. Impedance measurements seem to support these results. On the other hand, concerning the stability under simulated conditions of the intestine and gastric fluid, it was shown that the capsules that showed the best protection were P2.25I0.00A7.00, P3.00I0.51A2.38 (2.5x10^7, 1.5x10^7 respectively), and P0.00I1.50A7.00 (3.3x10^8 ). These data resulted interesting, since they are about three or four orders of magnitude larger when compared with other prepared formulas. In addition, they are about two orders of magnitude larger when compared with the control (1.3x10^5). Concerning the microstructure, it is found that the mixture with the largest viability show a microstructure simulating flakes with soft and well defined edges. Its appearance is slightly compacted favoring the release and protection of the probiotic bacteria. In conclusion, this research indicates that the use of probiotics and a polymeric matrix protect the viability of the Lactobacillus acidophilus against the stress conditionings, because of the micro-encapsulation process. They also protect its transit through the gastrointestinal tract. The results indicate that the bacteria are liberated in sufficient amount in a specific site (the colon), providing lots of benefits to the host.
