RESUMEN: Introducción. La obesidad se caracteriza por hipertrofia e hiperplasia del adipocito; ocasionada por el desequilibrio entre el consumo y el gasto energético. En México, el sobrepeso y la obesidad se presentan en el 70% de los adultos. La acumulación de grasa corporal tiene una base genética, no sólo en las formas monogénicas, sino también en la obesidad común. Por ello se han desarrollado diversas estrategias que permitan identificar los genes asociados al desarrollo de obesidad. Una de las más utilizadas es el estudio de genes candidatos que participan en procesos como la termogénesis, la adipogénesis y la lipólisis, tales estudios han sido realizados principalmente en poblaciones de origen caucásico. A la fecha, se conoce poco sobre la genética de la obesidad en la población mestiza y nativa mexicana. Por ello, el objetivo de este estudio fue valorar la contribución al desarrollo de obesidad en población mexicana de 15 polimorfismos de un solo nucleótido (SNP) ubicados en 13 genes ADRB2, ADRB3, ENPP1, LEPR, FABP2, IL6, UCP2, UCP3, RESISTINA, ADIPOQ, PPARG, GNB3, NR3C1. Materiales y Métodos. Se incluyeron 3,391 individuos mexicanos no relacionados, analizados en dos etapas: Etapa 1) Estudio de asociación caso-control, se analizaron los 15 SNPs de ADRB2 rs1042719, ADRB3 rs4994, ENPP1 rs1044498, LEPR rs1137101, FABP2 rs1799883, IL6 rs1800795, UCP2 rs659366, rs660339, UCP3 rs2075577, RESISTINA rs1862513, ADIPOQ rs2241766, PPARG rs3856806, GNB3 rs5443, NR3C1 rs56149945 en 873 adultos, incluyendo 570 obesos con un IMC ≥30 kg/m2 (de los cuales 189 eran obesos clase III) y 303 controles mayores de 30 años con un IMC
ABSTRACT: Introduction. Obesity is characterized by adipocyte hypertrophy and hyperplasia, caused by the imbalance between consumption and energy expenditure. In Mexico, overweight and obesity are present in 70% of adults. The accumulation of body fat has a genetic basis, not only in monogenic forms, but also in common obesity. Therefore it have been developed several strategies to identify genes associated with the development of obesity. One of the most used is the study of candidate genes involved in processes such as thermogenesis, adipogenesis and lipolysis; these studies have been conducted mainly in Caucasian populations. To date, few is known about the genetics of obesity in Mexican mestizo and native population.
