Las algas contienen varios fucanos y galactanos sulfatados que presentan actividad anticoagulante y es importante determinar si estas moléculas generan efectos secundarios a largo plazo. En el presente trabajo se evaluó la actividad moduladora de la coagulación sanguínea “in vivo e in vitro” de polisacáridos sulfatados obtenidos de Macrocystis pyrifera. El extracto crudo se obtuvo a partir de 1 kg de alga con agua destilada a 55°C. La solución clarificada fue sometida a un fraccionamiento sólido-líquido en DEAE-celulosa con eluyentes de NaCl a 0.5, 1 y 2 M. Se obtuvieron cuatro fracciones, de las cuales 06-019-61 F1A (23.529 g), mostró 183.90 en la prueba de tiempo de protrombina (TP) así como 260s de tiempo de tromboplastina parcial activada (TTPa). Se envió 0.2 g de la fracción a la Escuela Nacional de Medicina y Homeopatía-IPN para determinación de resonancia magnética nuclear y espectrofotometría de infrarrojo en CICIMAR-IPN. En las pruebas in vivo se aplicaron las dosis de 5, 2, 0.5 mg/kg de la fracción 06-019-61 F1A vía intraperitonial al organismo Mus musculus (Cepa CD-1) durante tres meses y se realizó análisis de biometría hemática, tiempos de coagulación (TP Y TTPa) y Química sanguínea (SMAC24). Cada uno de los análisis anteriores, se sometió al método estadístico de Friedman, el cual mostró diferencias significativas solo en dos parámetros de biometría hemática y un parámetro de SMAC 24. Los ensayos de TP y TTPa in vivo mostraron inhibición del coagulo en los primeros 15 días de tratamiento, después fueron normales (13s y 45s respectivamente). En los análisis histológicos se encontró infiltración bacteriana en ventrículo izquierdo de corazón, infiltración grasa en miocardio y pericardio; en hígado se encontró hemosiderina, hiperplasia y deformación de hepatocitos. En riñón se encontró deformación de túbulos proximales y distales, hiperplasia y dilatación glomerular. El pulmón mostró hiperplasia y al igual que el bazo, infiltración sanguínea y hemosiderina. En médula ósea se encontró infiltración grasa. Abstract
Algae contain various sulfated galactans and fucans having anticoagulant
activity and it is important to determine whether these molecules produce long term
side effects. In this study we evaluated the modulatory activity of blood coagulation "in
vivo and in vitro" sulfated polysaccharide obtained from Macrocystis pyrifera. The
crude extract was obtained from 1 kg of seaweed with distilled water at 55 ° C. The
clarified solution was subjected to a solid-liquid fractionation on DEAE cellulose
eluting with NaCl at 0.5, 1 and 2 M. Four fractions were obtained, of which F1A 06-

In vivo tests were applied doses of 5, 2, 0.5 mg/kg of fraction F1A 06-019-61 via
intraperitoneal at the organism Mus musculus (strain CD-1) for three months and
performed blood count analysis, clotting times (PT and aPTT) and blood chemistry
(SMAC24). Each of the above analyzes, underwent Friedman statistical method,
which showed significant differences in only two parameters OF blood count analysis
and only one in SMAC 24. The PT and aPTT tests showed inhibition of in vivo clot in
the first 15 days of treatment, were normal after (13s and 45s, respectively). The
histological analyzes showed bacterial infiltration in the left ventricle of heart and fatty
infiltration in myocardium and pericardium; hemosiderin found in liver, hyperplasia
and hepatocyte deformation. Was found deformation in proximal tubule and distal in
kidney, hyperplasia and dilation of glomerulus. Showed hyperplasia in lung and like
spleen, blood infiltration and hemosiderin. In bone marrow fat infiltration was found. |
