RESUMEN: La globulina 11S de amaranto es la más abundante proteína de almacenamiento en
semillas maduras y es ampliamente reconocido por su valor nutricional. En este
estudio, esta proteína se le ha insertado cuatro pares del péptido valina-tirosina en
cada una de las regiones variables, generando cinco proteínas modificadas. Cada
una de estas proteínas fueron expresadas en E. coli BL21-CodonPlus(DE3)-RIL en
el medio F8PW diseñado en este estudio para la producción de proteínas
recombinantes. Después de purificar estas proteínas fueron analizadas por
espectroscopía de fluorescencia y dicroísmo circular, así como su estabilidad a la
desnaturalización térmica y química. Las inserciones en las regiones variables
afectaron en la expresión de las proteínas observando un mayor rendimiento de la
proteína sin modificar. Las proteína modificadas mantienen la estructura Beta I
como la que presenta la proteína nativa, sin embargo, la modificación de la región
II, III, IV y V, modifica el patrón de replegamiento in vitro de la proteína nativa. Las
inserciones en las regiones III y V cambian la estabilidad térmica de la globulina 11S
de amaranto. La proteína R5 forma una estructura ordena diferente a la proteína
nativa y al ser digerida por proteasas digestivas liberan los péptidos bioactivos VY
presentando actividad funcional in vitro.
ABSTRACT: The amaranth 11S globulin is
the most abundant storage protein in mature seeds
and is widely recognized for its nutritional value. In this study, this protein has been
inserted four pairs of valine-tyrosine peptide in each of the variable regions,
constructing five modified proteins. Each of these proteins was expressed in E. coli
BL21-CodonPlus (DE3) –RIL strain in the F8PW medium which it was designed in
this study for the production of recombinant proteins. After purifying these proteins,
they were analyzed by fluorescence and circular dichroism spectroscopy, as well as
their stability to thermal and chemical denaturation. The bioactive peptide insertions
in the variable regions affected the expression of the proteins, observing a higher
yield of the unmodified protein. The modified proteins maintained the beta I structure
as the one presented by the native protein, however, the modification in the regions
II, III, IV and V, modifies the in vitro refolding pattern. The insertions in regions III
and V change the thermal stability of 11S amaranth globulin. The R5 protein
acquired an arranged structure different to the native protein and the bioactive
peptides presented in vitro functional activity after that protein were digested by
digestive proteases. |
