RESUMEN:
La incidencia de Fusarium verticillioides en el cultivo de maíz y su peligrosidad
en la salud humana, y animal ha llevado a la búsqueda y desarrollo de genotipos
tolerantes. La pared celular tiene un papel clave durante el ataque fúngico, y la
detección de genes, cuyos cambios en la expresión se asocien a la tolerancia o
susceptibilidad a Fusarium, pudiesen ser usados en el desarrollo de marcadores
moleculares, permitiendo rastrear estas características en menor tiempo para la
selección de nuevos genotipos. Por lo que el objetivo del proyecto fue determinar el
cambio en la expresión de los genes involucrados en la biosíntesis de la pared celular
de raíces en plantas de maíz tolerantes y susceptibles a fusariosis en respuesta a la
infección por F. verticillioides. Para ello se emplearon dos genotipos, uno tolerante
(Tol) y uno susceptible (Sus) con y sin infección por Fusarium verticillioides. Mediante
un experimento de curso temporal se identificó el comportamiento de los genotipos
infectados para la extracción del ARN y determinación histoquímica de fluorescencia
endógena en la raíz del maíz. Se secuenciaron tres bibliotecas por cada condición para
la identificación de los genes expresados diferencialmente. En el día siete se
observaron los primeros signos de fusariosis a nivel de raíz en el genotipo susceptible,
mientras que en genotipo tolerante hasta al día nueve. Considerando al día siete para
la toma de muestra de raíz para la extracción del ARN y determinación histoquímica,
debido a la presencia de fusariosis en mayor grado en el genotipo susceptible. Se
observó un incremento significativo en la fluorescencia endógena de la pared celular
en el genotipo susceptible durante la infección por Fusarium, mientras que el Tol no
presentó cambios. Por otro lado, se identificó un total de 338 (Tol) y 140 (Sus) GED,
de los cuales 31 y 17 están asociados la pared celular en Tol y Sus, respectivamente.
Tol presentó la inducción de genes asociadas a la respuesta contra el estrés
oxidativo, respuesta defensiva y reforzamiento de la pared celular. Mientras en Sus se
presentó la inducción de genes asociadas a la respuesta defensiva y reforzamiento.
Lo que sugiere que la pared celular tiene un papel importante durante el ataque del
patógeno
ABSTRACT:
The incidence of Fusarium verticillioides in the cultivation of maize and its danger
in human and animal health has led to the search and development of tolerant
genotypes. The cell wall plays a key role during the fungal attack, and the detection of
genes, whose changes in expression are associated with tolerance or susceptibility
to Fusarium, could be used in the development of molecular markers, allowing to trace
these characteristics in less time for the selection of new genotypes. So the aim of
