ABSTRACT: The in vitro, and keratinocyte (HaCAT cells) cultures expression of four putative genes encoding for secreted aspartyl proteases of Candida dubliniensis (CdSAP1-4) is reported for the first time. The regulation of the expression of CdSAP1 and CdSAP3 genes cloned and sequenced in this work and CdSAP2 and CdSAP4 previously deposited in GeneBank database was studied. Also CdSAP7, CdSAP8, CdSAP9, and CdSAP10, orthologous genes of Candida albicans, were recognized in C. dubliniensis genome. The CdSAP1 and CdSAP2 expressions were independent from the morphological stage of C. dubliniensis, they are expressed at both pH 4 and pH 7, and were induced with albumin as nitrogen source. CdSAP3 expression was regulated by the pH, and was related with the infection process of keratinocytes. Expression of CdSAP4 predominated during the mycelial phase and the initial stage of keratinocytes infection. During infection of the HaCaT cell line, only genes CdSAP3-4 were expressed, keratinocytes were affected in their number and shape by the infection with C. dubliniensis, however, this effect decreased in the presence of pepstatin (aspartyl proteases inhibitor). Based on the aforementioned, we suggest that the Saps from C. dubliniensis could be considered virulence factor just as those from C. albicans, and as participants in the nitrogen metabolism of the yeast for nutrient acquisition. A total Yeast-mycelium transition was observed in YCB-BSA and Lee-FCS media.
RESUMEN: En este trabajo se reporta por primera vez la expresión de cuatro genes putativos codificantes de aspartil proteasas secretadas de Candida dubliniensis (CdSAP1-4). Se clonaron los genes CdSAP1 y CdSAP3 utilizando iniciadores degenerados y junto con los genes reportados en las bases de datos (CdSAP2 y CdSAP4) se estudió la regulación de la expresión de estos cuatro genes a nivel transcripcional bajo diferentes condiciones in vitro. También fueron reconocidos en el genoma de C. dubliniensis los genes ortólogos de C. albicans CdSAP7, CdSAP8, CdSAP9, and CdSAP10. La expresión de los genes CdSAP1 y CdSAP2 fue independiente del estado morfológico de C. dubliniensis, se expresaron a pH 4 y pH 7 y fueron inducidos con albúmina como fuente de nitrógeno. La expresión de CdSAP3 fue regulada por el pH y se relacionó con el proceso de infección de queratinocitos. La expresión de CdSAP4 predominó durante la fase micelial y el estado inicial de la infección de queratinocitos. Durante la infección con C. dubliniensis de la línea celular de queratinocitos HaCaT, solo se expresaron los genes CdSAP3-4 y se afectó la forma y el número de los queratinocitos, sin embargo este efecto disminuyó en la presencia de pepstatina A (un inhibidor de aspartil proteasas). Por lo anterior se sugiere que las Saps de C. dubliniensis pudieran ser consideradas al igual que las de C. albicans atributos de virulencia así como participar en el metabolismo nitrogenado de la levadura para la adquisición de nutrientes.
