RESUMÉN: En México existen pocos estudios sobre el comportamiento de la cantidad de inóculo depositado de Escherichia coli O157:H7 y la interacción de factores del medio (pH, actividad de agua) donde se desarrolla el patógeno y ambientales (como la temperatura). Del entendimiento de la interacción de los diferentes factores que condicionan el desarrollo de un patógeno sobre un medio de cultivo se puede predecir el inicio de la fase lag, la tasa de crecimiento del mismo y de esta manera proponer medios de control o monitoreo de posibles contaminaciones. La información obtenida nos dará idea de las condiciones más adecuadas que dirijan a la inocuidad del alimento. Además al inicio de este estudio se trabajó con un indicador de contaminación fecal como E. coli ATCC 25922 (genérica o no patógena) para conocer su comportamiento y la interacción del microorganismo con factores como pH, actividad de agua (aw), temperatura y concentración del inóculo microbiano. Es por ello que se planteó el presente estudio cuyo objetivo fue determinar el comportamiento de Escherichia coli patógena y no patógena en caldo de cultivo mediante diferentes métodos y parámetros fisiológicos ajustando su comportamiento con modelos matemáticos. En el trabajo experimental se evaluó la dinámica de crecimiento de E. coli O157:H7 y E. coli genérica influenciada por la temperatura (T°) (15, 25, 35 °C), pH (4, 5.5, 7.0), actividad de agua (aw) (0.965, 0.981, 0.998) y concentración del microorganismo (4, 400, 40000 UFC). Para evaluar el comportamiento del crecimiento microbiano se utilizaron cuatro técnicas, tres de ellas (Bioscreen, Calorimetría Diferencial de Barrido (DSC) e Impedancia), como alternativas al Conteo en Placa (CP). Se evaluó como respuestas de crecimiento para E. coli O157:H7 el inicio de la fase lag (λ), la tasa de crecimiento específico (µmax), el tiempo de generación (tg) y el valor máximo alcanzado (A). Los resultados obtenidos con Bioscreen y CP se usaron para ajustarse a los modelos de Gompertz, Logístico y Richards. Se encontró que usando la técnica de CP para E. coli no patógena fueron factores altamente significativos (p=0.000000) el pH y la concentración del microorganismo, la temperatura (p=0.000032), excepto la aw (p=0.641835). La interacción de la aw con el inóculo inicial (p=0.000000) y la temperatura (p=0.035351) fueron significativos. La aw es un factor que no debe ignorarse al evaluar el comportamiento de E. coli no patógena. Para la E. coli O157:H7 usando CP, los factores estudiados mostrando efectos altamente significativos (p=0.000) para los términos lineales (L) y cuadráticos (Q) para el pH, y la temperatura (L). La concentración inicial de E. coli patógena mostró significancia en los términos L (0.033) y Q (0.003). Usando el Bioscreen, se mostró que fueron altamente significativos (p=0.000000) el pH y la temperatura, ambos en sus términos (L) y (Q), así como el inóculo inicial (L). De esta información se concluye la importancia que muestra el pH, la temperatura, la concentración del microorganismo al evaluar el comportamiento del E. coli O157:H7. Sin embargo las gráficas del efecto de la aw muestran diferencias significativas que coinciden con el análisis de E. coli no patógena. El inicio de la fase λ mostró valores variables para las técnicas de Bioscreen (5.9 a 67.44 h), Calorimetría Diferencial de Barrido (0.23 a 4.83 h) e Impedancia (4.55 a 56.95 h), excepto para Vaciado en Placa (donde no se detectó fase λ). La fase λ se prolongó cuando la combinación de las condiciones evaluadas fueron criticas (pH 5.5, 15 °C, 0.965 aw e inóculos de 4 UFC). Las condiciones óptimas de crecimiento de E. coli O157:H7 fueron a pH 7, 0.998 aw a 35 °C, independientemente de la concentración de inóculo evaluado. Así mismo independientemente de la temperatura, pH, aw y concentración del microorganismo, a pH 4.0 no se observó crecimiento de E. coli patógena y no patógena en el caldo de cultivo en las cuatro técnicas evaluadas. A pH 4.0 las cinéticas de sobrevivencia fueron detectadas a 15 y 25 °C, y las cinéticas de muerte a 35 °C. En general el tg fue más corto al incrementar la temperatura, evidenciando que mientras se acerque al óptimo de crecimiento, E. coli se multiplicará a mayor velocidad. Al incrementar la temperatura el tg disminuye y µmax, incrementa. El crecimiento máximo (A) por E. coli O157:H7 varió de 9.99 a 8.55 log10 UFC/mL usando CP y en el Bioscreen fue de 1.2 a 0.57 DO. El modelo que permitió el mejor ajuste de los datos experimentales usando las técnicas de CP y del Bioscreen fue el modelo de Richards (R2 =0.999 a 0.992) y el modelo Logístico (R2 =1.0 a 0.993), respectivamente. La Calorimetría Diferencial de Barrido mostró ser la técnica alternativa al recuento tradicional para evaluar el comportamiento de E. coli. Esta técnica es rápida, no laboriosa, requiere de pequeños volúmenes de muestra.
ABSTRACT: In Mexico doesn´t have been studied the behavior about the quantity of inoculation deposited of Escherichia coli O157:H7 and the interaction of environment, the culture (pH, water activity) and their factors where is developed the pathogen and its environment (like the temperature). The interaction among factors can condition the development of a pathogen in its environment and it predicts when the phase lag begins, the rate of growth, so we can control it or monitor it another ways of contamination, all of this help to us to be careful and the food can be inocuity .Also when this investigation began, it was used a indicator of fecal contamination (E. coli ATCC 25922) to know its behavior and the interaction of microorganism with factors like pH, water activity, (aw), temperature and concentration of inoculums microbial. That´s why this investigation was planned to know the behavior de Escherichia coli of pathogenic and non pathogenic in broth of culture with different methods and physiological parameters fitting its behavior with mathematics models. In the investigation it was evaluated the dynamic of growth of E. coli 0157:H7 influenced by the temperature (T) (15, 25, 35 °C), pH (4, 5,5, 7,0), water activity (aw) (0,965, 0,981, 0,998) and concentration of the microorganism (4, 400, 40000 UFC). In order to evaluate the behavior of the microbial growth four techniques were used, three of them (Bioscreen, Differential Scanning Calorimetric (DSC) and Impedance), like alternatives to Count in dishes (CP). It was evaluated as answers of growth for E. coli 0157:H7 beginning of phase lag (λ), the rate of specific (µmax), the time of generation (tg) and the reached maximum value (A). The results obtained with Bioscreen and CP was used to adjust the models of Gompertz´s, Logistic and Richards. It was used the technique of CP for E. coli non pathogen were factors highly significant (p=0.000000) pH and the concentration of the microorganism, the temperature (p=0.000032), except aw (p=0.6418335). The interaction of aw with inoculum initial (p=0.000000) and the temperature (p=0.035351) were significant. aw is a factor that must not ignore it when the E. coli´s behavior non pathogen is evaluated. For the investigation de E. coli 0157:H7 using CP, factors show highly significant effects (p=0.000) for the lineal (L) and quadratic (Q) terminus for pH, and the temperature (L). The first pathogen concentration of E. coli showed great significance in the terminus L (0.033) and Q the temperature, both of them in their terminus (L) and (Q), and the inoculums initial (L). vii This information shows the important of the pH, the temperature, and the concentration of microorganism when the behavior of E. coli 0157:H7 is evaluated, although the graphics of effect of aw show significant differences that they coincide with the analysis of E. coli non pathogen. The beginning of the phase λ showed variable values for the techniques of Bioscreen (5,9 to 67,44 h), Differential Scanning Calorimetric (0,23 to 4,83 h) and impedance (4,55 to 56,95 h), except for drained in plate (where it did not detect phase λ). The phase λ was prolonged when the combination of the conditions were evaluated and critical (pH 5.5, 15 °C, 0.965 aw and inoculums of 4 UFC). The optimal conditions of growth of E. coli 0157:H7 went to pH 7, 0.998 aw to 35 °C, it was independently of the concentration of inoculums evaluated, also independently of the temperature, pH, aw and concentration of the microorganism, pH 4,0 were not observed growth of E. coli pathogen and non pathogen in the broth of culture in the four techniques evaluated. About pH 4.0 the kinetics of survivor were detected to 15 and 25 °C and the kinetics of death to 35 °C. In general tg was shorter when the temperature was increasing, it approached the optimal one of growth, and E. coli was multiplied itself speeder. When the temperature increases, tg diminishes and µmax increases. The maximum growth (A) by E. coli 0157:H7 changed from 9.99 to 8.55 Log 10 UFC/mL using CP and in the Bioscreen was 1.2 to 0.57 DO. The Richard (R2 =0,999 to 0,992) model and the Logistic (R2 =1,0 to 0,993) model respectively were the best model that let the best adjust of the experimental dates, when it uses the techniques of CP and Bioscreen. Differential Scanning Calorimetric showed to be the best alternative to evaluate the behavior of E. coli, this technique is easy to use, and it requires a little bit of sample to work.
